I was waiting for a thread like this to be started. Desolation Row is by far his best work in terms of songwriting. People like Mr. Tambourine Man because of its thinly veiled drug referrences :P

But I think Tambourine Man is beautifully written, too.
I've seen Dylan in concert 13 times and I never get tired of him.

is what I voted for, it's the first song of his I heard as a kid that turned me on to his stuff, but the song that I was just listening to and really love is "When You Gonna Wake Up" off Slow Train Coming.

I LOVE the man and all his stuff, so it's hard for me to pick just one!!
