If he wanted, he could decline the offer. Of course, it's purely ceremonial unless there's a disaster that wipes out the POTUS, VP, and Speaker all at once.

The current President pro tempore of the Senate is Robert Byrd (D-West Virginia). He replaced Ted Stevens (R-Alaska), who held the office from January 3, 2003, to January 4, 2007. http://en.wikipedia.org/wiki/President_pro_tempore_of_the_United_States_Senate
