Does one have to have visited a country that they will be an ambassador to?

I have no idea what the answer is to this. Jon Stewart is making fun of the fact that Obama's ambassadors haven't been to the countries they are nominated for. I think that's a fair point, but should ambassadors have familiarity with the countries they are sent to? Should they have visited, lived, speak the language, or does it not matter at all?
