If Bob Mueller requested that grand jury investigation

be expanded, would Deputy AG Rosenstein agree or would he follow Trump's directive not to cooperate?

once he appointed Mueller with the mandate to investigate, he cant stop him short of removing him. Rosenstein actually answered this question recently saying that it was not a witchhunt and that mueller could investigate every and all matters related or not to the russian hack. Anything Mueller uncovers, he can pursue further.
